ABSTRACT

To study the chemical constituents and their biological activities in the rhizomes of Curcuma phaeocaulis, silica gel column chromatography, reverse medium pressure liquid chromatography, preparative thin layer chromatography, and semi-preparative high performance liquid chromatography were used for isolation and purification and modern spectroscopic methods were used to determine the structure of the isolated compound. Moreover, the effect of the compound on the proliferation of HUVECs was determined by the MTT assay. A new elemane-type sesquiterpenoid glycoside was isolated from the n-butanol soluble fraction of 95% ethanolic extract of the rhizomes of Curcuma phaeocaulis. Its structure was identified as (1Z)-2-hydroxy-curzerenone 2-O-β-D-glucoside. It showed no inhibitory effect on the proliferation of HUVECs.

ABSTRACT

@#Objective - To establish a new non exposed intratracheal instillation method for establishing a rat silicosis model. Methods , The specific pathogen free SD rats were randomly divided into control group and experimental group with ten rats in , each group. Rats in the control group were given 1.0 mL of 0.9% sodium chloride solution and rats in the experimental group - were given 1.0 mL of silica suspension with a mass concentration of 50 g/L adopting to the one time intratracheal instillation , - , method and then followed by ventilator assisted ventilation immediately. When the tidal volume stabilized at 2.0 mL the ventilator was removed and the tracheal intubation was pulled out. Five rats in each group were sacrificed after two and four , - Results weeks after modeling and hematoxylin eosin staining and Masson staining of lung tissue were performed. There was , , no death in the two groups of rats during the experiment. After two and four weeks the control group had normal lung structure , , , normal alveolar cavity size no inflammatory cell infiltration thin alveolar wall only a small amount of collagen distribution , around the lung interstitium and bronchus. At the second week of modeling the alveolar wall of the rats in the experimental , , , group was slightly thickened interstitial lymphocytes and macrophages were infiltrated slight hyperplasia was found and a , small amount of fibroblasts were visible. At the 4th week of modeling the alveolar wall of the rats in the experimental group was , , , , significantly thickened fibrous nodules were formed and fibroblasts fibrocytes collagen fibers were significantly increased. Conclusion - The combination of ventilator and non exposed intratracheal instillation method can be used to successfully , , . establish a rat silicosis model which is simple safe and effective

@#Abstract: Objective To analyze the serotype distribution, drug resistance rate and drug resistance gene carrying of Streptococcus pneumoniae isolates in hospitalized patients, and evaluate the coverage of the vaccine to the serotype of Streptococcus pneumoniae in this area, so as to provide reference for the rational use of antibiotics in clinic. Methods A total of 150 strains of non-repetitive Streptococcus pneumoniae isolated from inpatients from January 2015 to December 2019 were collected for serotyping and antimicrobial sensitivity test. The carrying rates of pbp2b, ermB and tetM were detected by PCR. Results The PCR classification rate of 150 strains of Streptococcus pneumoniae was 93.1%, and the classification rate of capsular swelling test was 100%, and a total of 19 serotypes were divided, mainly 19F and 6B. Children's serotypes were predominantly 19F, 6B, and 15A; adult serotypes were predominantly 19F, 14, and 23F. The coverage rates of the PCV7, PCV10, PCV13 and PPV23 vaccines were 36.8%, 42.1%, 57.9% and 68.4%, respectively. Strains with serotypes of 19F, 6B, 3, and 23F had higher rates of resistance to antimicrobials. The sensitivity of Streptococcus pneumoniae to penicillin was greater than 96.0%. Antimicrobials with significant differences in resistance rates between invasive and non-invasive strains were penicillin, moxifloxacin, and levofloxacin. The percentage of strains carrying both ermB and tetM resistance genes was 96.0%, and the concordance rate between pbp2b, ermB and tetM resistance genes and the resistance phenotype was >98.0%. A total of 10 multi-resistance combinations were detected, with a multi-resistance rate of 62.6%, and the multi-drug resistance pattern of Streptococcus pneumoniae was mainly concentrated in the 19F and 6B serotypes. Conclusion There are significant age differences in the serotypes of Streptococcus pneumoniae in this area. The vaccine currently used has low coverage in this region and therefore offer limited protection to the population. The drug resistance rates of Streptococcus pneumoniae varied significantly among serotypes. Erythromycin and tetracycline are not recommended for clinical treatment of Streptococcus pneumoniae. Penicillin can still be used as the first choice for clinical treatment of Streptococcus pneumoniae infection.

ABSTRACT

To investigate the chemical constituents of Anisodus tanguticus, silica gel column chromatography, Sephadex LH-20 column chromatography, preparative thin layer chromatography, and semi-preparative HPLC were used to separate and purify the chemical constituents from the extract of A. tanguticus. The planar structure of the isolated compound was identified by HRMS, IR, and 2D NMR experiments. The absolute configuration of the isolated compound was determined by a combination of NOESY, coupling constant, circular dichroism (CD), and transition metal chelate reagent dimolybdenum tetraacetate [Mo2(OAc)4]-induced circular dichroism (ICD) data analysis. A new compound of the anisotane-type sesquiterpene (1) was isolated, which was determined to be (1R,2S,3R,4R,6R,7R,9R)-anisotane-11(13)-ene-3,4,9-triol and named anisotanol F. This is the second report of anisotane-type sesquiterpene, which has previously been reported as a novel sesquiterpenoid skeleton by our research group. Furthermore, the cytotoxicity against HUVECs and inhibitory effect on NO release in LPS-induced RAW264.7 cells of compound 1 were investigated. However, the results showed that it was inactive. Compound 1 is a new compound isolated from A. tanguticus. It belongs to the unusual anisotane-type sesquiterpene. This result enriches the chemical composition of A. tanguticus.

ABSTRACT Introduction: Physical exercise can enhance the body's ability to adapt to the external environment and improve the contractility of the heart. At the same time, it can help improve blood circulation and increase lung capacity. Objective: This article explores the critical role of sports in the prevention of cardiovascular diseases. Methods: This article conducts tests on related physiological indicators such as hemodynamic rheology for students who regularly participate in physical exercise and those who do not participate in physical exercise. Results: Students who exercise regularly and those who do not exercise mobilize faster cardiovascular function at the beginning of work. This shows good adaptability to sports. Conclusion: The use of fitness exercise prescription by students who exercise comprehensively has the most pronounced effect on improving vascular function. Level of evidence II; Therapeutic studies - investigation of treatment results.


RESUMO Introdução: O exercício físico pode aumentar a capacidade do corpo de se adaptar ao meio ambiente e melhorar a contratilidade do coração. Ao mesmo tempo, pode reestabelecer a circulação sanguínea e aumentar a capacidade pulmonar. Objetivo: Este artigo explora o papel fundamental do esporte na prevenção de doenças cardiovasculares. Métodos: Este estudo conduziu testes em indicadores fisiológicos relacionados, tais como a reologia hemodinâmica em estudantes que regularmente praticam exercício físico e aqueles que não praticam exercício físico. Resultados: Estudantes que praticam exercícios regularmente e aqueles que não o fazem mobilizam uma função cardíaca mais rápida no início do treino, o que demonstra boa adaptabilidade ao esporte. Conclusão: A prescrição de exercícios de preparo físico para estudantes que se exercitam de forma completa tem o efeito mais pronunciado na função vascular. Al mismo tiempo, puede restablecer la circulación sanguínea y aumentar la capacidad pulmonaria. Objetivo: Este artículo explora el papel fundamental del deporte en la prevención de enfermedades cardiovasculares. Métodos: Este estudio condujo pruebas en indicadores fisiológicos relacionados, como la reología hemodinámica, en estudiantes que regularmente practican ejercicio físico y los que no lo practican. Resultados: Estudiantes que practican ejercicios regularmente y los que no lo hacen movilizan una función cardíaca más rápida al principio del entrenamiento, lo que demuestra buena adaptabilidad al deporte. Conclusión: La prescripción de ejercicios de preparación física para estudiantes que se ejercitan de forma completa tiene el efecto más pronunciado en la función vascular. ABSTRACT

By summarizing the existing researches on the prevention and treatment of damp-heat syndrome with traditional Chinese medicine(TCM), this paper digitized the TCM syndromes of damp-heat syndrome, enriched the TCM theories, and provided reference for the prevention and treatment of damp-heat syndrome and provided innovative ideas for the research of damp-heat syndrome. Damp-heat syndrome can cause pathological changes all over the body. It is easy to induce a variety of acute febrile diseases, including ulcerative colitis, chronic gastritis, chronic hepatitis B, chronic kidney disease, irritable bowel syndrome, pneumonia, cough and asthma, which seriously affects the health of the body. Damp-heat syndrome can be divided into syndrome of damp-heat in upper Jiao, syndrome of damp-heat in middle Jiao and syndrome of damp-heat in lower Jiao .At the same time it can be divided into damp-heat obstructing the lung, large intestine damp-heat, damp-heat containing the spleen, liver-gallbladder damp-heat, and bladder damp-heat syndrome , nephritic damp-heat syndrome by simulating clinical pathogeny according to basic theories of TCM. Heat-clearing and dampness-expelling is the treatment principle of damp-heat syndrome. TCM achieves the purpose of treating damp-heat syndrome by virtue of its anti-inflammation, anti-oxidation, regulation of intestinal flora, lowering blood glucose, lowering blood lipid and enhancing immunity. Based on the two aspects of TCM and prescription, the application, mainly in the syndrome differentiation of triple energizer differentiation, visceral syndrome differentiation is complementary, pharmacological action and mechanism of TCM in prevention and treatment of damp heat syndrome are reviewed in detail, which is a concrete analysis and discussion, in order to provide relevant experience for data integration and system construction of damp heat syndrome, and provide new ideas and help for follow-up clinical drug research and development and optimization innovation of damp heat syndrome. At present, the material basis and relevant prevention and treatment mechanism of dampness-heat syndrome are still not clear in China, so it is necessary to further study and explore its potential biological diagnostic markers, strengthen the research and development of new Chinese medicine preparation against dampness-heat, and elaborate the mechanism of action of Chinese medicine on damp-heat syndrome in multiple ways and directions.

Objective@#To investigate psychosexual health of students during pubertal transition in Taizhou and explore the influence of family factors.@*Methods@#A questionnaire survey was conducted among 1 215 students in senior grades of primary school, junior and senior high schools in Taizhou by stratified cluster sampling.@*Results@#The average score of adolescents sexual and mental health questionnaire was (3.82±0.52), the sexual knowledge score was (3.31±1.00) and the score of sexual adaptation was (4.29±0.68). Sexual attitude and sexual control among female students were significantly better than male students (t=4.53,6.96,P<0.01). The four dimensions of physiological knowledge, sexual knowledge, sexual concept and self-adaptation made significant progress with the improvement increased with educational background(F=157.18,85.18,85.96,65.54,P<0.01). The parent-child relationship, parental sex education, the only child in the family, education background of parents, family economic status and the long-term separation from parents in early childhood had predictive effects on the development of puberty students’ psychosexual health (P<0.05).@*Conclusion@#Students during pubertal transition in Taizhou show moderate level of psychosexual health. Schools should strengthen sexual knowledge education.In order to promote students’ psychosexual health, more attention should be paid to the influence of family factors.

OBJECTIVE@#To evaluate the clinical efficacy of umbilical moxibustion in the treatment of patients with idiopathic pulmonary interstitial fibrosis complicated with gastroesophageal reflux of lung-spleen deficiency.@*METHODS@#A total of 116 patients with idiopathic pulmonary fibrosis complicated with gastroesophageal reflux of lung spleen-deficiency were randomized into an observation group (58 cases) and a control group (58 cases). In the control group, the routine treatment was applied; in the observation group, on the base of the treatment as the control group, the umbilical moxibustion was used. The treatment was given 2 times a week, 2 h each time for 12 weeks, and followed up for 12 weeks after treatment. The composite physiological indicators (CPI), TCM syndrome scores, the St. George respiratory questionnaire score (SGRQ) and 6-minute walk test (6MWT) were compared before and after treatment in the two groups.@*RESULTS@#The improvement of the TCM syndrome scores (cough, post-activity shortness of breath, fatigue, heartburn, acid reflux), CPI score and 6MWT in the observation group was better than those in the control group (all 0.05). There was no significant difference in the scores of CPI scores and syndromes (cough and fatigue) between the two groups at 12 weeks after treatment (all >0.05). The 6MWT, SGRQ scale scores and syndrome scores (post-activity shortness of breath, acid reflux, heartburn) were better than those in the control group in the follow up (all <0.05).@*CONCLUSION@#Umbilical moxibustion can improve the ventilation and diffusion function of lung in patients with idiopathic pulmonary fibrosis and gastroesophageal reflux of lung-spleen deficiency, reduce clinical symptoms and improve patients' quality of life and activity endurance. And it has long-term effect.

ABSTRACT CONTEXT: Inflammatory myofibroblastic tumors are a rare type of soft-tissue tumor. Inflammatory myofibroblastic tumors are characterized by rearrangements involving the anaplastic lymphoma kinase gene locus on 2p23. Case Report: We report the case of a 67-year-old Chinese male who presented with dysuria and fever. Magnetic resonance imaging showed an irregular prostatic mass with an isointense signal and obscure boundary. Histopathological evaluation showed that the mass consisted mainly of spindle-shaped cells. Immunohistochemical evaluation showed that the tumor cells were negative for anaplastic lymphoma kinase. CONCLUSIONS: Inflammatory myofibroblastic prostate tumors are rare lesions with unclear etiology. The pathological diagnosis is very important.

ABSTRACT

A series of berberine derivatives were synthesized by introducing substituted benzyl groups at C-9. All these synthesized compounds (4a-4m) were screened for their in vitro antibacterial activity against four Gram-positive bacteria and four Gram-negative bacteria and evaluated for their antifungal activity against three pathogenic fungal strains. All these compounds displayed good antibacterial and antifungal activities, compared to reference drugs including Ciprofloxacin and Fluconazole; Compounds 4f, 4g, and 4l showed the highest antibacterial and antifungal activities. Moreover, all the synthesized compounds were docked into topoisomerase II-DNA complex, which is a crucial drug target for the treatment of microbial infections. Docking results showed that H-bond, π-π stacked, π-cationic, and π-anionic interactions were responsible for the strong binding of the compounds with the target protein-DNA complex.

ABSTRACT

Objective To investigate the distribution of insulin resistance (HOMA-IR) index and its influencing factorsamong middle and old aged people with normal glucose and to provide the basis for early screening and prevention of type 2diabetes. Methods A total of 229 residents were selected with health records showed normal blood glucose (fasting bloodglucose < 7.0mmol/L, postprandial 2h blood glucose<11.1 mmol/L) and more than 40 years old from July, 2012 to June,2015. Height, weight, waist and hip circumference, and the fasting plasma glucose (FPG), insulin (FINS), lowdensity lipoprotein (LDL), uric acid, tumor necrosis factor (TNF) and interleukin -6 (IL-6) were recorded to analyzethe distribution of HOMA-IR and its influencing factors. Results Totally 229 people were included, of which 113 were male(49.34%), 116 female(50.66%) . The average age was(63.58 + 8.85) years old. The average HOMA-IR index was 0.94(1.08) and there were 21 people that HOMA-IR exceed the standard (HOMA-IR≥2.68), accounting for 9.17%.TheHOMA-IR index of different gender, age, waist circumference, hip circumference, uric acid in the elderly had significantdifference (P < 0.05) .Multiple linear regression analysis showed that HOMA-IR index was positively correlated withfemale, waist circumference and IL-6 and was negatively correlated with age. Conclusion The possibility of IR was higherin women with relatively low age, female, central obesity and high IL-6 levels among the middle and old aged people withnormal blood glucose.

objective To explore the role of Xanthatin in the targets of epithelial-mesenchymal transition (EMT) process using molecular docking method,and the effect on the target protein expression of HepG2 cells was detected by Western assay.Method Dhs,Vimentin,Snail and VEGFR3 are critical targets in EMT process,the spatial binding ability of Xanthium was evaluated by molecular docking method,compared with the corresponding endogenous substances:nicotinarnide adenine dinucleotide,Acetate ion,flavin adenine dinucleotide,and N-Acetyl-D-glucosamine.HepG2 cells were cultured,and the effects of Xanthatin of 1,5 and 20 mol/L concentrations on Dhs,Vimentin,Snail and VEGFR3 protein expression were detected by Western Blotting assay.Rusult Molecular docking show that Xanthatin has obvious affinity to key factors of EMT process such as Dhs,Vimentin,and VEGF-R3,higher than that of endogenous substance;and the affinity with Vimentin was less than that of endogenous substance;Western Blotting experiments proved the virtual results.The expression of Vimentin,Snail,VEGFR3 protein was significantly lowered,and the expression of e-cadherin was significantly raised.Conclusion The influence of Xanthatin to key factor e-cadherin,Vimentin,Snail,VEGFR3 are obvious,Which is likely to be a potential target.The results of computer virtual experiment and Western Blotting have certain similarity.Molecular virtual docking can pre hint the potential target factor.

Objective To establish a method for determining the contents of polyphyllin Ⅰ and polyphyllin Ⅱ in Kangkeling Mixture. Methods The contents of polyphyllin Ⅰ and Ⅱ were determined by HPLC gradient elution. Poroshell 120 Ec-C18 column (4.6 mm × 150 mm, 4 μm) was used; Acetonitrile-water (A:B) was set as the mobile phase; the flow rate was 1.0 mL/min; the detection wavelength was 210 nm; column temperature was 30 ℃. Results Polyphyllin Ⅰ showed good linear relationship in the range of 1.009–10.09 μg (r=0.999 6), and the average recovery was 97.31% (RSD=2.05%, n=6). Polyphyllin Ⅱ showed good linear relationship in the range of 0.640 5–6.405 μg (r=0.999 8), and the average recovery was 96.41% (RSD=1.67%, n=6). Conclusion The method is simple, with good repeatability and accurate results, which can be used to determine the contents of polyphyllin Ⅰ and Ⅱ in Kangkeling Mixture.

ABSTRACT

Objective To establish a method for determining the contents of polyphyllin Ⅰ and polyphyllin Ⅱ in Kangkeling Mixture. Methods The contents of polyphyllin Ⅰ and Ⅱ were determined by HPLC gradient elution. Poroshell 120 Ec-C18 column (4.6 mm × 150 mm, 4 μm) was used; Acetonitrile-water (A:B) was set as the mobile phase; the flow rate was 1.0 mL/min; the detection wavelength was 210 nm; column temperature was 30 ℃. Results Polyphyllin Ⅰ showed good linear relationship in the range of 1.009–10.09 μg (r=0.999 6), and the average recovery was 97.31% (RSD=2.05%, n=6). Polyphyllin Ⅱ showed good linear relationship in the range of 0.640 5–6.405 μg (r=0.999 8), and the average recovery was 96.41% (RSD=1.67%, n=6). Conclusion The method is simple, with good repeatability and accurate results, which can be used to determine the contents of polyphyllin Ⅰ and Ⅱ in Kangkeling Mixture.

ABSTRACT

This study employed a Bac-to-Bac/Bombyx mori bioreactor to mass-produce immunogenic urease subunit B (UreB) from Helicobacter pylori. The signal peptide bombyxin from B. mori was used to promote secretory expression to improve expression levels and was designed and integrated into the UreB gene to generate the Bacmid/BmNPV/(signal peptide)-UreB baculovirus expression system. To determine whether the bombyxin signal peptide resulted in secretory expression of recombinant UreB (rUreB) and to determine the secretory efficiency, we tested the secretory expression level of rUreB in Bm5 cells using ELISA. To further investigate whether secretory expression affected cell viability, cells were evaluated using 0.4% trypan blue staining, and Bacmid/BmNPV/UreB without the signal peptide served as a control. The above recombinant bacmid constructs were injected to silkworm larvae, and the secretory expression level of rUreB was detected using SDS-PAGE and semi-quantitative western blot analysis. The results indicated that the bombyxin signal peptide directed the secretory expression of rUreB and that this expression improved the viability of Bm5 cells. Moreover, the results showed that the expression level of rUreB was 1.5 times higher with the Bacmid/BmNPV constructs containing the bombyxin signal sequence than those without the signal sequence. These results demonstrate that secretory expression can enhance rUreB expression levels and is likely to aid in the large-scale expression and yield of rUreB in silkworm larvae.

ABSTRACT

Objective: To study the low polar chemical constituents from Leonurus japonicus. Methods: The chemical constituents were isolated and purified by various chromatographic methods including column chromatography over silica, Sephadex LH-20, and Rp C18 columns. Their structures were identified by physicochemical properties and spectroscopic analysis. Results: Six aliphatic compounds were isolated from the EtOAc extracts of L. japonicus, including two allenes: (-)-nonadeca-5,6-dienoic acid methyl ester (1) and methyl octadeca-5,6-dienoate (2); four saturated aliphatic compounds: heneicosanoic acid (3), arachidic acid (4), heptacosanoic acid (5), and methyl myristate (6). Conclusion: All the compounds are isolated from L. japonicus for the first time. Compound 1 is a new one named leonuallenote A, while compound 2 is first reported from the plants of Leonurus L.

ABSTRACT

To study the changes of pupil diameter and its effects during phacoemulsification. METHODS: Block design, prospective study. 60 cataract patients (60 eyes) were selected, which divided into two groups according to the age, young age group (group A, 20 eyes of 20 patients) and older age group (group B, 40 eyes of 40 patients). Two groups were all given compound tropicamide eye drops for fully mydriasis before operation, computer software to process images during operation, and according to the cornea amplification ratio measure pupil diameter during the operation phase: before the corneal tunnel incision ( t1 ) , after injection of sodium hyalurantae ( t2 ) , before artificial lens implantation (t3), at the end of the surgery ( t4 ). Statistical analysis of data using SPSS18. 0 software.RESULTS: The pupil diameter measurements of t1-t4 were followed respectively:group A were 8. 36 ± 0. 65, 8. 97±0. 50, 8. 67±0. 63, 8. 72±0. 96mm; group B were 7. 73± 0. 58, 8. 23 ± 0. 59, 7. 89 ± 0. 16, 7. 70 ± 0. 63mm. Overall comparisons between the two groups, the pupil diameter measurements had differences during the operation phase ( F = 26. 696, P0. 05). Comparisons in a group at different time points: the pupil diameter of t2 was larger than that in t1, with statistically differences ( P>0. 05), the pupil diameter of t3 and t4 were decreased in group B, with statistically differences (P<0. 05).CONCLUSlON: Compound tropicamide eye drops are given for mydriasis before operation, young patients with cataract and senile patients with cataract can achieve the effect of surgery for mydriasis, the pupil stability of phacoemulsification in senile patients with cataract are lower than that in young patients with cataract.
